※細(xì)胞數(shù)量為推薦鋪板細(xì)胞數(shù)量,需根據(jù)細(xì)胞大小調(diào)整鋪板數(shù)量(以感染時(shí)細(xì)胞密度30%為宜),按照實(shí)際感染時(shí)細(xì)胞數(shù)量去計(jì)算病毒加入量。
※為增加病毒與細(xì)胞接觸的幾率,可適當(dāng)減少感染時(shí)培養(yǎng)基的體積。
感染復(fù)數(shù)(Multiplicity of Infection, MOI),含義為感染時(shí)病毒和細(xì)胞數(shù)量的比值,即平均每個(gè)細(xì)胞感染的病毒活性單位數(shù)(慢病毒單位為TU/cell)。
實(shí)驗(yàn)中按照設(shè)置好的MOI計(jì)算每孔需要加入的病毒量:
每孔加病毒量(μl)=MOI×感染時(shí)的細(xì)胞數(shù)/滴度(TU/ml)×1000。
